LSK114 and FLO-MIN114

Dear ONT developing team. Maybe my experience with LSK114 will be helpful. I am sequencing gDNA of a protist. The problem of this DNA is that it blocks pores super rapidly (no matter what protocols I try). So, I usually run 2-3 sequencings on the same flowcell. LSK110 was alright (max throughput 1-1,5 GB from 3 runs on one flowcell). LSK112 was not: Lower throughput, bad pore recovery after DNAse treatment. LSK114 is just a monster:D I did 4 runs on the same flowcell - each run almost as good as the first one - and collected all in all 3 Gbp of data. I attach here the pore occupancy in two pictures: of the 1st run and of the 3 subsequent runs in a separate graph. However, it seemed that addition of BSA makes the buffer viscous and it does not flow well. I had to suck the buffer out though the waste pores to make the flush buffer and library enter the cell (for the 2d, 3d, and 4th loadings). Also, when compared to LSK110, LSK112 and 114 showed more adapters in the pore occupancy profile. Best, Victoria Shabardina
